Research overview

PNC-27 is a chimeric peptide, listed as a p53-derived and HDM-2-binding peptide, that joins a p53 sequence to a membrane-associating leader. It is used to study p53-related signalling and peptide-induced membrane disruption in cultured cells. Supplied at 5mg as a lyophilised powder for in-vitro laboratory research only.

The p53 region in this peptide is the part that binds HDM-2, also called MDM2. The attached leader sequence is described as helping the peptide enter or sit in cell membranes. In the literature, PNC-27 is reported to form pores in the membranes of some transformed cell lines by binding HDM-2 present at the cell surface, causing lysis, while non-transformed lines were reported to be spared. This model of membrane-surface HDM-2 is contested, and independent replication has been limited.

Laboratory work includes cell viability and lactate dehydrogenase release assays to detect membrane lysis, imaging of HDM-2 localisation, co-culture of transformed and non-transformed lines, and peptide-lipid interaction studies with model membranes. Control peptides with scrambled or leader-only sequences are standard. It is supplied as a lyophilised powder in a 5mg vial.

Questions

What are the two parts of PNC-27?

One part is a p53-derived sequence that binds HDM-2. The other is a leader sequence described as helping the peptide associate with cell membranes. Control peptides lacking either part are used to test which element is responsible for an effect.

How is PNC-27 supplied and stored?

It is supplied as a lyophilised powder in a 5mg vial. Keep it cold, dry and away from light, and reconstitute with a suitable laboratory diluent only when the material is needed.

Is PNC-27 a medicine?

No. It is supplied strictly for in-vitro laboratory research. It is not a medicine, food or supplement, and it is not for human or animal use. Buyers must be aged 18 or over.
